| Overview | The MILLIPLEX® MAP Human Neurological Disorders Magnetic Bead Panel 3 (Cat. No. HND3MAG-39K) is to be used for the simultaneous quantification of the following 7 analytes in any combination in CSF, serum or plasma samples: • Angiotensinogen (AGT) • Contactin-1 • Fetuin A • Kallikrein-6 • Osteopontin (OPN) • Soluble Superoxide Dismutase 1 (SOD1) • Soluble Superoxide Dismutase 2 (SOD2) NOTE: For serum and plasma samples Fetuin A requires a higher (1:4000) dilution and therefore must be assayed alone as a singleplex. For serum or plasma samples all analytes, except Fetuin A, require a 1:50 dilution and may be assayed together as a multiplex. |
| Background Information | Neurological disorders are diseases of the brain, spinal cord and nerves. There are more than 600 diseases of the nervous system, such as Multiple sclerosis (MS), Alzheimer's disease (AD), amyotrophic lateral sclerosis (ALS), Parkinson's disease (PD), and stroke as well as frontotemporal dementia and many others. Identification of biomarkers for these neurological disorders allows scientists to identify early signs of neurological damage, track disease progression, and monitor efficacy of therapeutic treatment. The MILLIPLEX® MAP 7-plex Human Neurological Disorders Magnetic Bead Panel 3 is designed for the simultaneous measurement of multiple biomarkers in human cerebrospinal fluid (CSF), plasma, and serum using the Luminex xMAP® instrument platform. The kit contains enough material to analyze 38 samples in duplicate, including the standard curve and two quality controls. NOTE: For serum and plasma samples Fetuin A requires a higher (1:4000) dilution and therefore must be assayed alone as a singleplex. For serum or plasma samples all analytes, except Fetuin A, require a 1:50 dilution and may be assayed together as a multiplex. |

| Application Notes | • This assay requires 25 µL diluted sample per well, sample dilution recommendations are as follows: o CSF samples: 1:100 dilution in kit Assay Buffer for all analytes. o Serum and Plasma samples: 1:50 dilution in kit Assay Buffer for all analytes except Fetuin A. NOTE: Fetuin A cannot be assayed along with other analytes in serum/plasma samples. Fetuin A should be diluted 1:4000 in the kit Assay Buffer and analyzed as a singleplex for serum and plasma samples. • An overnight (4°C) incubation is recommended (a 2 hour room temperature protocol has also been validated and is included in the protocol) |
